Born in Philadelphia in 1936, DePreist studied composition under Vincent Persichetti at the Philadelphia Conservatory of Music before earning both bachelor's and master's degrees from the University of Pennsylvania.

He was the nephew of the pioneering contralto Marian Anderson, whom DePreist acknowledged as an extraordinary role model.
In a 2005 interview with NPR's Roy Hurst, he remarked, “She knew that she was Marian Anderson, but my aunt was simultaneously the most humble person I ever met in my life and the most powerful. It was a combination of her not needing to strut her strength because it was just a natural part of her. To the extent that anything has rubbed off, I'm grateful.”
Like his aunt, DePreist became a trailblazer, gaining recognition as one of the few African-American classical conductors during his time. Despite facing significant physical challenges, he won the admiration of orchestras and audiences around the world.
