The planned closure of the facility in Maesteg is part of wider plans to develop a new health and wellbeing centre in Bridgend's Llynfi Valley.
Protesters were joined on Saturday by deputy First Minister Huw Irranca-Davies, there in his capacity as local MS for Ogmore.
Mr Irranca-Davies, whose local surgery is based at the site, said staff and clinicians must be involved in decisions about local services.
Maesteg Community Hospital opened in 1914 to care for injured soldiers returning from World War I and was built thanks to the local miners of the Llynfi Valley who donated a penny a week for its construction.
Protesters said vital local services including a GP surgery, X-ray and mental health runs from the site, among other departments.
