Obligada a enfrentar revelaciones sobre su pasado, forja una relación con tres jóvenes destinadas a futuros importantes (interpretadas por Sydney Sweeney, Isabela Merced y Celeste O'Connor); pero antes, todas deben sobrevivir a un presente mortal amenazadas por el villano Ezekiel Sims. Madame Web (Dakota Johnson) podría servir como una especie de precuela o un juego multiversal, pues Julia Carpenter, Anya Corazón y Mattie Franklin en algún momento son Spider-Woman.

Es una anciana ciega que sufre de miastenia gravis, un trastorno autoinmune neuromuscular que la obliga a usar un sistema de soporte vital similar a una telaraña; de ahí su apodo (y el eslógan de la cinta: "Su red las conecta a todas"). Se dedica a guiar a los demás, por lo que no lucha activamente contra los villanos.
